Engineer for the analysis of visual pathologies
Context
The “Identification of molecular pathways involved in eye diseases, from gene to therapy” team, led by C. ZEITZ and I. AUDO, is setting up translational projects to study retinal diseases of genetic origin. AUDO, implements translational studies of retinal diseases of genetic origin: identification of mutations in patients, creation and characterization of in vivo models reproducing retinal diseases, mechanistic studies of retinal function and development of therapeutic protocols.
The proposed project involves studying the function of retinal genes associated with myopia using functional techniques (phenotyping by electroretinography, multiple electrode array, histology, behavioral tests, induction and measurement of myopia) in genetically modified mouse models. On the other hand, it will be necessary to set up strategies to rescue the phenotype observed by administration of therapeutic molecules or gene therapy in order to better understand the mechanisms involved in myopia and to propose proofs of concept of innovative therapies for its treatment.
Missions
The candidate will work in close collaboration with researchers, thesis students and engineers involved in these projects. He/she will participate in each of these stages:
- Genotyping and maintenance of mouse lines
- Animal handling for functional and behavioral studies
- Ocular phenotyping (behavior, function, structure) of transgenic animals to better understand molecules involved in retinal pathologies in vivo and ex vivo
- Application of pharmacological treatments
- Harvesting retinal tissue for histology and functional analysis
- Surgical techniques: implantation of myopia glasses, intracardiac injections
- Electrophysiological recordings on isolated retina with data analysis
- Formatting of results for scientific articles
Skills required
Knowledge:
- Basic programming language (Python) for electrophysiology data analysis and statistics
- Basic knowledge of small animal surgical techniques (Certificate of training in experimental small animal surgery would be a plus)
- Theoretical knowledge of retinal structure, function and basic genetic concepts
- Knowledge of pharmacology
- Ability to understand spoken English

Education and experience
- Minimum diploma required: Bac +3
- Management and handling of murine lines (minimum training certificate in animal experimentation, experimenter level)
- Dissection and preparation of tissues for post-mortem analysis
- Proficiency in basic molecular biology experimental techniques (genotyping, Western blot, RT-qPCR, immunohistochemistry, in situ hybridization, microscopy)
- Reading of scientific protocols in English
